Objective
This proposal aims to investigate the molecular genetic mechanisms responsible for the development of the vertebrate forebrain and head. The main objectives of the project are to determine the roles of ANF and Otx class homeobox genes in the development of the vertebrate head. The first vertebrate representatives of the ANF and Otx gene families were isolated in mouse and frog in the laboratories of the participants of this project (Simeone et al., 1992; Zaraisky et al., 1992). Members of these gene families are amongst the earliest genes known to be expressed in the forebrain primordium and are therefore likely to be involved in the initial steps of head and forebrain patterning.
The following specific issues will be addressed in the project:
the roles of ANF and Otx related genes will be investigated by misexpression studies in frogs and fish using normal proteins and chimaeric proteins which strongly repress transcription;
the regulation of XANF-1 transcription will be investigated through functional analysis of the promoter region of XANF-1 gene;
the transcriptional regulation and possible functional roles of ZANF and Otx1-3 in zebrafish will be investigated by examining mutant strains of zebrafish in which regulation of these genes is perturbed;
possible regulatory interactions between ANF and Otx genes will be investigated by misexpression studies in both zebrafish and frogs;
regional patterning of the forebrain will be investigated in zebrafish through a combination of detailed comparative expression analyses of ANF and Otx genes with other regionally expressed forebrain markers together with analysis of mutations in which forebrain patterning is perturbed.
The results obtained will be published in the leading scientific journals and reported at the international conferences.
