Objective
TO PRODUCE CELLULASES, HEMICELLULASES, PECTINASES, AMYLASES AND PROTEASES BY SOLID-STATE FERMENTATION OF WASTES USING RELEVANT FUNGAL STRAINS.
A - CURRENTLY AVAILABLE STRAINS OF TALAROMYCES EMERSONII, TRICHODERMA REESEI, TRICHODERMA KONINGII, PENICILLIUM CAPSULATUM, PENICILLIUM PINOPHILUM, FUSARIUM SOLANI AND NEOCALLIMASTIX FRONTALIS WILL BE GROWN BY SOLID-STATE FERMENTATION ON A VARIETY OF AGRICULTURAL AND FOREST WASTES AND THE EXTRACELLULAR ENZYME SYSTEMS PRODUCED BY EACH STRAIN ON EACH SUBSTRATE IDENTIFIED.
B - NEW STRAINS OF MICROORGANISMS CAPABLE OF FERMENTING THE ABOVE WASTES WILL BE ISOLATED BY TRADITIONAL MEANS AND BY NEWER PROCEDURES INCLUDING PROTOPLAST FUSION OF GENETICALLY SIMILAR OF ORGANISMS (EG P.PINOPHILUM AND P.CAPSULATUM). THE COMPLEMENT OF ENZYMES PRODUCED BY SUCH STRAINS WILL BE IDENTIFIED AS ABOVE.
C - PROCEDURES FOR SOLID-STATE FERMENTATION OF WASTES BY SINGLE STRAINS OR BY APPROPRIATE COMBINATIONS OF STRAINS WILL BE OPTIMISED AS WILL PROCEDURES FOR EXTRACTION OF THE ENZYMES. INDIVIDUAL ENZYMES AND "COKTAILS" THEREOF WILL BE PREPARED.
D - BECAUSE OF THEIR HETEROGENOUS NATURE, THE WASTES TO BE USED AS SUBSTRATES WILL, AS HAS BEEN SHOWN, INDUCE THE SYNTHESIS OF CELLULASES, HEMICELLULASES, PECTINASES AND PROTEASES. THE AMOUNTS OF EACH OF THESE ENZYMES PRODUCED WILL BE DETERMINED AND THEIR SUITABILITY WITH RESPECT TO INDUSTRIAL APPLICATION WILL BE EVALUATED. THIS WILL INVOLVE EXAMINATION OF SUBSTRATE SPECIFICITY, PH AND TEMPERATURE OPTIMA, STABILITY UNDER CONDITIONS OF APPLICATION, SENSITIVITY TO INHIBITION AND EASE OF RECOVERY ETC...
E - APPROPRIATE PROCESSES DEVELOPED AT THE LABORATORY-SCALE WILL BE SCALED UP TO PILOT-PLANT LEVEL.
