Objective
The work aims at the development and construction of advanced laser desorption ionization mass spectrometry instrumentation with special emphasis on the analytical demands of molecular biology research and biotechnology.
The instrumental set-up will be modular with a basic configuration for routine molecular mass screening of biopolymers and mixtures thereof by matrix-assisted laser desorption ionization (MALDI) with high accuracy and sensitivity. Upgrading comprises an ion reflectron for improved mass resolution and mass accuracy emphasizing the capability to perform structural analysis of the analytes by analysis of metastable fragment ions (PSD : post-source-decay analysis) e.g. for peptide sequencing and glycoconjugate investigation. Upgrading furthermore includes the implementation of appropriate sample stages in order to couple MALDI-MS to routine liquid separation techniques, especially to (micro-)HPLC and capillary electrophoresis. Within the project, procedural protocols are to be developed both to improve the current performance of the MALDI technique with respect to reproducibility of preparations including tolerance against contaminants and shot-to-shot fluctuations and to adapt these protocols to the requirements of samples eluted from liquid chromatography.
