I) BIOTECHNOLOGICAL PREATREATMENTS : FULL DELIGNIFICATION
II) TRANSFORMATION OF BYPRODUCTS (AROMATICS) TO USEFUL COMPOUNDS
1. SCREENING OF ADDITIONAL STRONGLY LIGNOLYTIC MICROORGANISMS FROM EITHER SOIL SAMPLES FROM EXTREME BIOTOPES (WOODS,TROPICS ETC.) OR CULTURE COLLECTIONS LIKE ATCC,CBS,DSM. THE EXPERIENCE WITH CELLULOLYTIC ORGANISMS SHOWS THAT THE NATURAL VARIABILITY WITHIN SPECIES AND STRAINS IS RATHER HIGH. CONSIDERING THE SMALL NUMBER OF KNOWN ACTIVE LIGNOLYTIC ORGANISMS, CHANCES ARE RATHER HIGH TO FIND BETTER STRAINS.METHODS: RADIORESPIROMETRIC DETECTION FIRST, THEN FURTHER CHARACTERISATION BY THE LISSAMIN-LIGNIN-METHOD AND OTHERS.

ELUCIDATION OF THE POSSIBLE TRANSFORMATION OF LIGNIN-OLIGOMERS TO PHENOLIC COMPOUNDS OF COMMERCIAL VALUE, EITHER SPECIAL CHEMICALS LIKE VANILLINE OR MASS CHEMICALS (LIKE BINDING SUBSTANCES IN INDUSTRIAL PROCESSES).METHODS: PRESCREENING WITH RADIOLABELLED COMPOUNDS,SUBSEQUENT ANALYSIS WITH HPLC OR TEST FOR BINDING CAPABILITY.
3. EVALUATION OF POSSIBLE GENETIC MANIPULATIONS (MUTAGENESIS OR GENETIC ENGINEERING) FOR OBTAINING FUNGAL STRAINS WITH HIGH ACTIVITY IN LIGNIN (AND PHENOLICS) TRANSFORMATION BUT LOW DEGRADATION ACTIVITY. THESE STRAINS SHOULD BE OPTIMAL TRANSFORMERS OF AROMATICS.
4. ELUCIDATION OF THE XYLAN-DEGRADING ENZYME COMPLEX IN FUNGI WHICH ARE ENHANCED IN THEIR GROWTH WITH XYLANE. ISOLATION OF "XYLANASES" FOR THE ENZYMATIC HYDROLYSIS OF XYLOSES WHICH ARE BYPRODUCTS OF THE "ORGANOSOLV"- AND STEAM-EXTRACTION-PROCESSES.
5. ANALYSIS OF LIGNIN-OLIGOMERS WHICH ARE BYPRODUCTS OF THE "ORGANOSOLV"- AND STEAM-EXTRACTION PROCESSES AND ELUCIDATION OF THEIR POSSIBLE TRANSFORMATION BY APPROPRIATE STRAINS OF MICROORGANISMS.
6. OPTIMIZATION OF THE BIOTECHNOLOGICAL PROCESSES WHICH HAVE BEEN FOUND TO BE APPLICABLE UP TO FERMENTOR-SCALE.
7. ELABORATION OF PROCEDURES AND POSSIBLE INDUSTRIAL PROCESSES FOR THE USE OF LIGNIN-BYPRODUCTS OF THE ORGANOSOLV AND STEAM-EXTRACTION PROCESSES AS USEFUL INDUSTRIAL MASS PRODUCTS, ESPECIALLY AS BINDING MATERIALS IN THE ENZYMATIC GLUING-PROCEDURE.
8. SYNTHESIS AND PROCESSING OF SPECIFICALLY RADIOACTIVE AND CHEMICALLY LABELLED SUSBTANCES (14 DEGREE-AND LISSAMINE-TAGGED) AND POLYMERS, INCLUDING MATERIAL FROM LABELLED PLANTS,PARTIALLY DONE IN THE ZENTRALES ISOTOPEN LABORATORIUM DER UNIVERSITAET GOETTINGEN.
