USE OF MYCORRHIZAS IN POPLAR MANAGEMENT

Objective

THE AIM IS TO EVALUATE THE MYCORRHIZAL DEPENDENCE OF THE POPLAR AND TO UNDERSTAND THE RELATIONSHIPS BETWEEN ECTOMYCORRHIZAL AND VESICULAR-ARBUSCULAR MYCORRHIZAL INFECTIONS.

POPLAR SEEDLINGS OF SELECTED CLONES OBTAINED BY IN VITRO MICROPROPAGATION WILL BE PLANTED IN POTS CONTAINING STERILIZED SOIL AND THEN INOCULATED WITH DIFFERENT ECTOMYCORRHIZAL TUBER SPECIES, USING SPORES OR MYCELIA AS INOCULUM. ALTERNATIVELY SEEDLINGS WILL BE INOCULATED WITH SPORES OF DIFFERENT VAM FUNGI OR WITH ALREADY ESTABLISHED VAM ROOTS.
IN ORDER TO EVALUATE THE RELATIONSHIPS BETWEEN ECTOMYCORRHIZAL AND VAM INFECTIONS, IN OTHER EXPERIMENTS SEEDLINGS WILL BE INOCULATED WITH BOTH TYPES OF SYMBIONTS. ALL THE SEEDLINGS WILL BE KEPT IN A CONSTANT ENVIRONMENT ROOM, WATERED DAILY WITH DISTILLED WATER AND FED WITH A NUTRIENT SOLUTION. AFTER 3-6 MONTHS, THE INFECTION WILL BE ESTIMATED.
THE FOLLOWING FEATURES WILL BE CONSIDERED BOTH IN MYCORRHIZAL AND NON MYCORRHIZAL PLANTS:

A) SHOOT HEIGHT, LEAF DEVELOPMENT;

B) DRY WEIGHT OF THE SHOOT AND ROOT;

C) CONTENT IN LIGNIN, CELLULOSE, HEMICELLULOSE IN THE SHOOT;

D) MORPHOLOGICAL DESCRIPTIONS OF THE DIFFERENT TYPES OF INFECTIONS;

E) ESTIMATION OF THE INFECTION INTENSITY.
