Objective
FIRST THE SPECIES AND THEN THE METHODS ARE DESCRIBED.
-PSEUDOTSUGA. THE RESEARCH ON MICROPROPAGATION WAS DIRECTED TOWARDS PHOTOSYNTHESIS, TOPOPHYSIS AND ARCHITECTURE. BUDS WERE GROWN IN VITRO, ELONGATED AND ROOTED; HOWEVER, THE EFFICIENCY IS STILL LOW. THE BEST RESULTS WERE OBTAINED WITH THE SO-CALLED SEED PROPAGATION: MICROPROPAGATION STARTING WITH BUDS WHICH WERE INDUCED ON EMBRYOS OR YOUNG SEEDLINGS. THE METHOD HAS TO BE OPTIMIZED.
ALSO FUNDAMENTAL RESEARCH IS NEEDED. SEED PROPAGATION IS IMPORTANT BECAUSE THE AVAILABILITY OF SEED FROM SUPERIOR TREES IN THE COMING YEARS IS A SOURCE OF SERIOUS DOUBT.
THE METHOD IS ALSO IMPORTANT FOR THE SEED ORCHARD PROGRAM.
-QUERCUS. SEED PRODUCTION IS IRREGULAR; CONSERVATION IS IMPOSSIBLE. SEED PROPAGATION WILL PREVENT IMPORTS OF SEEDS OF DOUBTFUL ORIGINS. OTHER METHODS OF MICROPROPAGATION WILL SPEED UP TREE IMPROVEMENT.
-ALNUS. ALNUS TREES ARE BECOMING INCREASINGLY IMPORTANT FOR ENERGY PRODUCTION AND SOIL IMPROVEMENT (N-FERTILIZATION).
MICROPROPAGATION WILL REDUCE THE NEED TO MASS PROPAGATE SUPERIOR GENOTYPES.
-SALIX. METHODS OF MICROPROPAGATION ARE NEEDED TO MASS PRODUCE NEW DISEASE FREE CLONES OF SALIX SPECIES.
-POPULUS, ULMUS AND PLATANUS. THE METHODS THAT WERE DEVELOPED NEED FURTHER OPTIMIZATION.
-MICROPROPAGATION FROM BUDS; LEAF DEVELOPMENT, ELONGATION AND ROOTING IN VITRO. FOR SOME FOREST TREE SPECIES, IT HAS NOT YET BEEN DEVELOPED; FOR OTHERS ITS EFFICIENCY IS STILL LOW. MAJOR ATTENTION WILL BE PAID TO THIS METHOD.
-MICROPROPAGATION FROM EMBRYOS (SEED PROPAGATION). IN MOST TREE SPECIES THE AVAILABILITY OF SUPERIOR SEEDS IS STILL VERY LIMITED. EMBRYOS FROM THESE SEEDS CAN BE CULTURED IN VITRO AND CAN BE STRESSED TO FORM NUMEROUS PLANTLETS PER EXPLANT. AT THIS MOMENT THE METHOD IS ONLY USED FOR A FEW SPECIES.
-MICROPROPAGATION FROM MERISTEMS. PRODUCTION OF TREES FROM MERISTEM CULTURES RESULTS IN HEALTHY MATERIAL. SO FAR ONLY SOME BROADLEAVED SPECIES PRODUCED FROM MERISTEMS.
MICROCUTTINGS IN CULTURE TUBES: TRANSPORT OF MICROPROPAGATED MATERIAL. IN THE PAST, SOME DISEASES HAVE BEEN TRANSFERRED TO OTHER COUNTRIES WITH THE TRANSPORT OF PLANT MATERIAL. TRANSPORT OF TREES AS DISEASE FREE MICROCUTTINGS IN CULTURE TUBES ENABLES SCIENTISTS TO EXCHANGE MATERIAL WITHOUT RISKS. THE METHOD HAS TO BE OPTIMIZED TO REDUCE THE RISKS OF LOSS OF REGENERABILITY DURING TRANSPORT.
-MICROPROPAGATION FROM CALLUS OR CELL SUSPENSIONS. SO FAR ONLY ULMUS AND POPULUS WERE PRODUCED FROM CALLUS CULTURES. THE MORE FUNDAMENTAL ASPECTS OF THIS TECHNIQUE HAVE TO BE STUDIED BEFORE IT CAN BE APPLIED TO OTHER SPECIES.
ATTENTION WILL BE PAID TO THE REGENERATION PROCESS. THE OTHER METHODS HAVE A HIGHER PRIORITY SINCE THE RISKS OF GENETIC ABERRATIONS ARE FEWER.
-REALIZATION OF A GENE BANK OF MICROCUTTINGS. IT IS POSSIBLE TO CONSERVE PLANT MATERIAL IN CULTURE TUBES FOR VERY LONG PERIODS WHEN THE PHYSICAL CIRCUMSTANCES ARE MODIFIED. .
