Objective
Protease (PR) inhibitors have dramatically improved AIDS therapy, but resistant variants emerge even under patent combination therapy. Monitoring and overcoming PR inhibitor resistance therefore is of utmost importance for long-term control of HIV infection. This collaborative project will analyse resistance against HIV PR inhibitors in a European multimember study, provide a rational basis for understanding resistance development and viral evolution on a molecular level, and design and test novel compounds to overcome resistance. Closely collaborating groups from five European countries with long experience on biochemical, structural and biological aspects of HIV PR and its inhibitors, and clinicians treating AIDS patients will join in this interdisciplinary project, directly addressing, ant microbial drug resistance and changes in virulence', the defining topic of thematic priority 2.2.2 of the QOL programme.
