Engineering of the cellular quality control systems in bacillus subtilis for the production of high value-added proteins

Objective

The Bacillus protein secretion machine contains cellular quality control systems that efficiently remove misfolded or incompletely synthesised proteins. Paradoxically, these quality control systems represent a major problem for the production of heterologous proteins at commercially significant concentrations. The ultimate objective is to develop a set of super-producer strains. Full use will be made of recent advances in genomics (eg.: DNA arrays), proteomics, bioinformatics, directed molecular evolution and information obtained from previous, EU-funded programmes on Bacillus protein secretion, genome sequencing and the systematic gene fuction analysis. The proposal includes the potential to 'spin-off a research-based biotechnology SME or trust company.
