Objective
We have previously successfully generated biosynthetic heparin from E. coli K5 polysaccharide using enzymatic and chemical modifications. We now want to identify new regio- specific heparan sulphate structures by screening biosynthetic ologasaccharide libraries with FGF4, FGF8 and HIV-Tat protein. Chemical and enzymatic processes will be used to generate these oligasaccharides in vitro from the K5 polysaccharide. Furthermore, both yeast and mammanlian cells, which genetically alter their biosynthetic capacity, will be used to synthesize neoGAGs. The biological efficacy of these oligosaccharides will be screened in tumour growth assays to demonstrate their antitumor activity. This can lead to new antitumor therapies. Similarly, also the HIV-Tat protein recognizing oligosaccharides will be tested for their ability to modulate the biological activity of extracellular Tat.
The expert system is a windows based software which supports Experts and Non Experts during the feasibility analysis of Large Scale Solar System integration into. Default value setting is provided by databases, nomogram algorithms and calculation models. Especially Non Experts are support by the expert system through project specific default value setting. Only a view parameters are required as an input and all other parameters for system analysis are identified by the expert system.
