Objective
This project will produce Cell Factories (CAS) for the clean synthesis of antibiotics (presently produced via costly and environmentally unfriendly procedures), production of templates for modification into antibiotics/beta-lactamase inhibitors and which will make possible the commercial production of broad-spectrum beta-lactamase inhibitors. The CAS will utilise modified versions of natural enzymes involved in beta-lactic biosynthesis. The work will involve implementation of five Work Packages forming an integrated program with detailed structural, bioinorganic, engineering and fermentation studies on the relevant enzymes. The project will also produce a structural database for ferrous dependent oxygenases, define rules for engineering their selectivity and develop new methodology for crystallography and mutant screening.
