Nicotine, nicotinic receptors and ageing

Objective

The project regroups the laboratories of M. Zola (I), D. Rubinsztein (UK), J.P. Changeup (F), K. Fuse (S), P. Inciter (D) and P. Moser (Synthelabo Recherché, F). Its aim is to explore the role of nicotinic acetylcholine receptors (nAChR) in brain ageing and related deficits. The workplan includes:
1. To study,whether polymorphisms in nAChR genes influence the rate of cognitive decline in the elderly;
2. To characterise the molecular mechanisms of nicotine neuroprotection and neurodegeneration caused by nAChR deficits in animal models, including the study of
a. the influence of nacho isotopes on brain ageing using nacho subunit mutant mice,
b. the interaction between nacho deficits and other age-related pathogenic factors (e.g. amyloidal beta protein and tropic factors),
c. the influence of nacho deficits on neuronal apoptosis, and d. the comparative genomic of ageing in humans and nacho mutant mice;
3. To characterise new, nicotinic agents for the therapy of age-related neurodegeneration and cognitive deficits.
