A diagnostic test for abnormal prion protein in live animals based on fluorescent immunoassay using capillary electrophoresis and hplc

Objective

As a result of the BSE crisis in Europe, a number of diagnostic tests have been developed for the detection of abnormal prion protein, the putative agent of Transmissible Spongiform Encelopathies. A major problem in the study of TSE is that there are no tests able to detect the abnormal prion protein in the early stages of the disease. The aim of this proposal is to further improve and validate a method based on competitive immunoassay involving fluorescent-labelled peptides analysed by capillary electrophoresis (CE) and micro-High Performance Liquid Chromatography (µHPLC). This method which has been mainly used to detect PrPsc in sheep blood samples will be extended to the detection of bovine prions and to the pre-symptomatic detection of PrPsc in humans, from the analysis of blood and other biological samples such as cerebrospinal fluid (CSF) and urine.
