ENZYMATIC CONVERSION OF THE CARBOHYDRATES OF STRAW INTO SOLUBLE SUGARS.

Objective



A NUMBER OF FUNGI ARE KNOWN TO HAVE THE ABILITY TO HYDROLYSE THE CELLULOSE AND HEMICELLULOSE CONSTITUENTS PRESENT IN CEREAL STRAWS AND SIMILAR AGRICULTURAL WASTES. ENZYME PREPARATIONS FROM THE FUNGI CAN BE USED TO CONVERT STRAW TO SOLUBLE SUGARS, WHICH COULD THEN FORM THE BASIS FOR PRODUCTION OF FUEL, SINGLE CELL PROTEIN, ETC.
TO IMPROVE THE EFFICIENCY OF SUCH CONVERSIONS ONE NEEDS TO OBTAIN STRAINS OF THE FUNGI SHOWING CERTAIN ADVANTAGES OVER THE WILD-TYPE, VIZ. PRODUCING THE POLYSACCHARIDE-HYDROLYSING ENZYMES
A) MORE ABUNDANTLY
B) IN THE ABSENCE OF THEIR INSOLUBLE SUBSTRATES
C) SO ALTERED THAT THEY ARE NOT INHIBITED BY THE END-PRODUCTS OF THEIR ACTIVITY
SCREENING METHODS ARE PROPOSED FOR THE SELECTION OF MUTANTS WITH THESE CHARACTERISTICS, USING CULTURE ON AGAR PLATES. THEY WILL BE APPLIED TO VARIOUS SPECIES OF STRAW DECOMPOSING FUNGI AND TO MUTANT STRAINS OF TRICHODERMA REESEI ALREADY AVAILABLE.
SELECTED STRAINS WILL BE TESTED FOR THEIR ABILITY TO PRODUCE ENZYMES IN SUBMERGED CULTURE. ENZYME PREPARATIONS SO OBTAINED WILL BE TESTED ON STRAWS CHEMICALLY PRETREATED TO INCREASE THEIR SUSCEPTIBILITY TO ENZYME ATTACK.
