Evaluation of microbiological methods for detection and for enumeration of microbiological contaminants in foods

Objective

At the moment, ISO microbiological methods have not been subjected to inter laboratory tests.
The acceptance of these methods by CEN requires this confirmation.

The objective of the proposal is to determine precision data (expressed in terms of repeatability and reproducibility) for methods which have been identified as a priority.
The final validation will countain a definition of repeatability and reproducibility and the precision limits that analysts should expect to achieve when using the methods.
As soon as they are collected and interpreted, results will regularly be sent to CEN for information. Once approved by CEN, the precision parameters will be incorporated in the relevant CEN standards.
In that way, further barriers to international trade should be removed;
The first trial in progress concerns Bacillus cereus.
The results of the pretrial have shown a good homogeneity and stability whichever the type of sample. A small problem of transport observed in one case should be solved for the trial proper. Final result will be discussed on December 16th in Brussels.
The second trial in progress (Listeria monocytogenes : enumeration and detection) has been discussed during a meeting held in Norwich, 26th June 1997, and the pretrial will begin at the end of October. Statistical tests to be applied to qualitative data will be extensively reviewed and discussed on December 15th.
The trials concern methods which have been identified as a priority, i.e. the enumeration of Bacillus cereus (ISO 7932), Staphylococcus aureus (ISO 6888), Clostridium perfringens (ISO 7937), Listeria monocytogenes (ISO / DIS 11292-2),and also the detection of Salmonella (ISO 6579) and Listeria monocytogenes (ISO / DIS 11290 1) as specified in the dedicated call (95 / C 14806).
For each type of micro-organism, the performance of each method will be determined using samples of meat, cheese, and a dried food.
A pretrial comprising the participation of three expert laboratories, will be carried out prior to each formal trial to provide estimations of precision and ensult protocol is satisfacting.
Participants have been selected who are accreditated to either the EN 45000 series or to ISO 9000, or who are at least likely to achieve formal accreditation in the very near future - 20 to 25 participants will be involved in each trial.
Tripartite agreement exists between IDF, AOAC and ISO in order to facilitate full international agreement on the methods. Each trial will be carried out according to the internationally harmonized protocol.
