Objetivo
The aim of this proposal is to develop reference methods to detect and quantify the presence of non-milk proteins in milk powder. Taking into account the availability, cost per kg and organoleptic properties, the detection of non-milk proteins will be restricted to the detection of protein isolates from soya, wheat, maize and pea. In addition the detection of some enzyme treated soya proteins will be evaluated.
Genetic varieties and a multiplicity of extraction and isolation schemes, including enzyme treatment, complicates the detection and quantitation of plant proteins. This proposal envisions the testing and evaluation of fundamentally different analytical methods in order to find at least two reference methods capable of detecting protein isolates of soya, wheat, maize and pea. The availability of two methods (or more) increases the chance to detect other genetic varieties of plant protein and to have success with other dairy products. Preferably, the reference method should be able to detect simultaneously the presence of these proteins, if not, then additional reference methods will be evaluated to enable the detection of all four plant proteins mentioned.
The project starts with the collection of representative samples of plant protein isolates and genuine milk powder (low and high heat). Evaluated will be an immunological method (ELISA), a combination of size separation and (if necessary) immunologic detection (SDS-PAGE immunoblotting) and two separation methods (RP-HPLC and CZE). If necessary the selectivity in HPLC will be enhanced by the use of on-line API-MS (single ion monitoring). Enzymatic N-deglycosylation will be studied in combination with RP-HPLC and CZE in order to reduce multiplicity of peaks and/or peak broadening.
Alternatively, the separation (anion-exchange HPLC or CZE of derivatives) of the liberated N-linked oligosaccharides might be evaluated to detect the presence of plant proteins. Methods should afford detection limits < 5% m/m of plant protein (protein/protein) and preferably as low as 1%.
Finally, two or more methods will be chosen for a collaborative study, which involves the participation of several subcontractors from various EU-member states. The results will be disseminated in the Official Journal of the EU, international journals and on symposia.
