Objectif
Background Currently, a scientific authoritative, harmonised European scheme is not available for detecting and diagnosing Xiphinema americanum-group nematode species which are recognised as being quarantine pests by both the European Union (Commission Directive 77/93/EEC, amended annex I/A1) and the European and Mediterranean Plant Protection Organisation (EPPO) (A2 Quarantine List No. 150). Several members of the X. americanum group are of phytosanitary significance as they are efficient vectors of four important plant viruses of quarantine significance: cherry rasp leaf nepovirus (EU annex I/A1; EPPO A1/127); peach rosette mosaic nepovirus (EU annex I/A1; under consideration by EPPO); tobacco ring spot nepovirus, potato calico strain (=potato black ringspot virus, EU annex I/A1; EPPO A1/128) and tomato ring spot nepovirus (EU annex I/A1; EPPO A2/102).
The four viruses have been subject to pest risk analysis and are considered to present the risk of serious economic impact in Europe. The virus-vector potential of X. americanum- group species indigenous and widespread in the EU region has not been determined. However, the risks presented by the nematode species occurring outside the region are that they provide a means of transporting the viruses to the region and could become established and disseminate the viruses. Objectives The project will produce an internationally authoritative protocol for detecting and identifying Xiphinema americanum-group virus-vector nematode species for incorporation into the set of diagnostic protocols on quarantine pests being developed by the EU.
Work programme: The meet this objective the partners in the project will:
1) rationalise the 44 putative morphospecies in the X. americanum-group;
2) evaluate the virus-vector abilities of the most common species;
3) produce a definitive list of X. americanum-group species based on best fit morphological and genetic data;
4) establish a genomic DNA library from which molecular probes with X. americanum-group/species specificity will be identified;
5) screen labelled-lectins and produce and screen a panel of monoclonal antibodies prepared against X. americanum-group nematodes to identify those exhibiting X. americanum-group/species specificity;
6) develop a detection method for recovering X. americanum-group specimens directly from mixtures of soil nematodes involving genus/species specific lectins and monoclonal antibodies bound to paramagnetic beads;
7) integrate the results from the above to provide a practical, reliable and cost effective protocol for the detection and diagnosis of virus-vector X. americanum-group nematodes;
8) co-ordinate and analyse an evaluation of the protocol, based on ring tests; and finally;
9) establish a definitive scheme for the detection and diagnosis of virus-vector X. americanum-group nematode species for inclusion in the set of diagnostic protocols on quarantine pests being developed by the EU.
State of progress: The project started on 1 October 1998. The partners had a first meeting during the Nematology symposium at Dundee (early August).
