Objectif
The general objectives of this project are as follows:
- Build a reference collection of probiotic strains used in feeds - Gather together existing methods and results and check for their applicability
- Develop/adapt methods for the enumeration of probiotics in feeds - Develop/adapt methods for the identification at species level of probiotics used in feeds (cultural/biochemical)
- Develop/adapt methods for the differentiation of strains used as probiotics
- Validate these methods by collaborative studies within the European Union
- Draft standard methods for approval by the EU
The proposal outlines the issues involved arising from Council Directive 70/524/EECconcerning additives in feeds, specifically probiotics. Little information exists in the literature on analytic methods for probiotics in animal feeds and what methods there are have been proposed by manufacturers. Iew of the latter have been subjected to ring testing.
This proposal deals with a project that intends to collate the information available on the proposed methods of analysis and develop and adapt the best ones available for eventual interlaboratory comparisons.
Detection and enumeration will be investigated using traditional culture based techniques and species level identification will be made using both traditional biochemical test profiles and, where feasable, polymerase chain reaction using species-specific primers. Characterisation of strains within species will be carried out using advance molecular techniques such as pulsed field gel electropheresis and RAPD-PCR. In this way it will be possible to compare original industrial strains with those isolated from a product sample.
The work will be done by five partners chosen from different member states for their experience with particular organisms, techniques or animal feeds. Each partner will work on a particular organism and the methods for its detection, enumeration and strain characterisation.
Animal feed samples will be prepared by industrial collaborators on a good will basis and these samples will be used for development and adaptation of methods.
The results of this work will not only lead to the existence of harmonised methods for control of probiotics in animal feeds but also to the promotion of trade within the community, benefits to the livestock industry and eventually to the consumer.
