TRANSMISSION OF MALARIA PARASITES FROM MAN TO MOSQUITO REQUIRES THE PRESENCE OF SEXUAL STAGES IN THE BLOOD BUT THE PROCESS IS DETERMINED BY COMPLICATED AND ENIGMATIC FACTORS. AIM OF THE SEXUAL STAGE VACCINE IS BLOCAGE OF TRANSMISSION, AND KNOWLEDGE ABOUT THE VARIOUS FACTORS HELPS ITS DEVELOPMENT. INFECTIVITY OF GAMETOCYTES IN THE FIELD CANNOT DIRECTLY BE ESTIMATED USING THE PARASITIC DEVELOPMENT BOTH BY FACTORS FROM THE HUMAN HOST AND THE MOSQUITO VECTOR. THE PROJECT APPROACHES TRANSMISSION MECHANISTICS OF PLASMODUM FALCIPARUM IN CAMEROON FROM THE EXPERIMENTAL SIDE AS WELL AS EPIDIOMINOLOGICALLY. THE PRESENCE OF TRANSMISS. REDUCING FACTORS IS DETECTED BY PERFORMING EXPERIMENTAL FEEDINGS OF BLOOD FROM GAMETOCYTE CARRIERS TO LOCALLY REARED ANOPHELINES. THE NATURE OF THESE FACTORS, BE THEY PHASE PROTEINS AND THEIR EFFECTS ALONG THE SPOROGONIC DEVELPMENT WILL BE ANALISED.
THE PRESENCE OF TRANSMISSION
BLOCKAGE IS ALSO DETECTED IN HUMAN POPULATIONS UNDER VARIOUS DEGREES OF EXPOSURE TO INFECTED MOSQUITOS THROUGHOUT THE YEARLY CYCLE OF SEASONS, IN AGE GROUPS AND IN HEALTHY AND SICK PEOPLE. NEWLY DEVELOPED ELISAS FOR ANTIBODIES AGAINST 45/48 AND 230 KD GAMETE PROTEINS WILL BE INTRODUCED AND APPLIED LOCALLY, WHILST A BIO ASSAY WITH CULTURED GAMETOCYTES AND MOSQUITOES IS AT HAND TO EXTEND THE DATA FROM THE FIELD.
THE PROJECT IS A CONCERTED ACTION OF THE UNIVERSITY NIJMEGEN ORSTOM PARIS AND OCEAC YAOUNDE THE REQUESTED DUTCH HALFTIME POSTDOC AND THE AFRICAN PHYSICIAN WILL CARRY OUT MOST OF THEIR RESEARCH IN CAMEROON FOR A PERIOD OF THREE YEARS.
