Design of light-harvesting DNA-nanoprobes with ratiometric signal amplification for fluorescence imaging of live cells.

Project description

Nanoscale RNA sensors for diagnostics

RNA-based methods are gaining momentum in the detection of viruses, the diagnosis of neurodegenerative diseases and tumour profiling. In addition, RNA monitoring is integral to many experimental processes as it impacts cell signalling, gene expression and cell survival. However, analysis requires cell manipulation and RNA extraction, which may cause RNA degradation and lead to erroneous results. The EU-funded DNANanoProbes project has developed DNA nanosensors capable of detecting RNA molecules by imaging. The technology employs fluorescent nanoparticles that hybridise with target RNA molecules. Amplification of signal leads to high sensitivity, which can be exploited for the diagnostic localisation and quantification of RNA in living cells.

Objective

The project is aimed to develop the novel technique of fluorescent signal amplification upon detection of RNA sequences in living cells up to a single copy level. The novel interdisciplinary approach will be focused on design of innovative fluorescent nanosensors based on polymer nanoparticles (NPs) incorporated with organic dyes, functionalized with DNA shell. The project is predicated on the innovative design of DNA-nanosensors, proposed by the Applicant, which is built on groundbreaking findings of the Host laboratory on ultrabright organic NPs and giant light-harvesting phenomenon that enable strong amplification in detection of nucleic acids. The project relies on contemporary spectroscopy/microscopy facilities and developments of the Host group, and the strong experience of the Applicant in synthesis of fluorescent dyes, design of DNA-probes and bioconjugation techniques.
