Unleashing arene oxidation via engineered unspecific peroxygenases
Direct epoxidation of arenes by cytochrome P450 is an important metabolic pathway in eukaryotes. The resulting arene oxides are also versatile precursors to many other classes of molecules. Despite this reaction’s tremendous potential for biotechnology applications, its replication has been elusive, hampering its exploitation. With the support of the Marie Skłodowska-Curie Actions programme, the UPOxArene project aims to achieve the epoxidation of arenes in a single step using engineered unspecific peroxygenases (UPOs). The researchers also plan to expand UPO chemistry beyond the aromatic hydroxylation accessible with cytochrome P450s by leveraging UPOs’ compatibility with organic media. The approach’s biotechnological relevance will be highlighted through production of industrially relevant compounds in continuous chemo-enzymatic cascades.
Objective
UPOXARENE is an interdisciplinary project combining organic synthesis, biocatalysis, and protein engineering, to realise the elusive epoxidation of arenes in a single step using engineered unspecific peroxygenases (UPOs). The compatibility of UPOs with organic media will be exploited to expand their chemistry beyond the aromatic hydroxylation accessible with cytochrome P450s. The utility of the method will be demonstrated through the preparation of industrially relevant products in continuous chemo-enzymatic cascades, in alignment with the principles set out in the MSCA Green Charter. UPOXARENE will promote biotechnology through the citizen-science initiative Peroxynauts, encouraging students from all backgrounds to pursue careers in the life sciences.
