Immunothrombolysis: deCoding neutrophiL Engagement in Active thrombus Resolution

Objective

Thrombosis remains a leading global cause of morbidity and mortality. In this process inflammation has been recognised as central driver besides platelets and coagulation. My work and that of others has defined thromboinflammation, the paradigm in which neutrophil–coagulation crosstalk drives vascular occlusions in the microvasculature and macrovasculature.

Recently, I introduced a previously unrecognized and paradigm-shifting concept: Neutrophils do not cease to act once a thrombus is formed. Instead, they continuously infiltrate established clots and acquire thrombolytic properties, directly promoting thrombus-resolution. I have termed this novel process immunothrombolysis, representing the conceptual opposite to thromboinflammation. A neutrophil-driven program of endogenous thrombus-clearance challenges one of the most entrenched assumptions in vascular biology. This has profound implications: If the underlying cellular, molecular and temporal mechanisms of immunothrombolysis can be harnessed, the immune-balance could artificially be tipped early towards endogenous thrombus resolution.
I hypothesize that neutrophil–coagulation interplay exerts stage-dependent, temporo-dynamically opposing effects, culminating in immunothrombolysis. To establish this new principle, I will:
1) Map the spatio-temporo-dynamic trajectories of continuous neutrophil infiltration, identifying the time and site where the switch from thromboinflammation to immunothrombolysis
2) Define the molecular triggers that reprogram neutrophils into a thrombolytic phenotype driving immunothrombolysis in the macrovasculature
3) Identify the molecular switches that induce endogenous microvascular thrombo-resolution to allow microvascular recanalyzation

This project could redefine thrombosis biology and pioneer endogenous immune-modulation for thrombus resolution, thereby reducing systemic therapeutic bleeding risks.

Fields of science (EuroSciVoc)

CORDIS classifies projects with EuroSciVoc, a multilingual taxonomy of fields of science, through a semi-automatic process based on NLP techniques. See: The European Science Vocabulary.

Keywords

Project’s keywords as indicated by the project coordinator. Not to be confused with the EuroSciVoc taxonomy (Fields of science)

Programme(s)

Multi-annual funding programmes that define the EU’s priorities for research and innovation.

Topic(s)

Calls for proposals are divided into topics. A topic defines a specific subject or area for which applicants can submit proposals. The description of a topic comprises its specific scope and the expected impact of the funded project.

Funding Scheme

Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

Call for proposal

Procedure for inviting applicants to submit project proposals, with the aim of receiving EU funding.

Net EU contribution

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
