Objective
The aim of this proposal is to enhance the European leadership in three-dimensional (3D) electron microscopy. HT-3DEM proposes to develop an automated platform to enable high throughput screening and analysis of native protein complexes and protein crysta ls by electron microscopy. For this purpose, HT-3DEM brings together European experts in electron microscopy, 2D crystallisation, machine vision, and robotics with leading manufacturers of electron microscopes and microsystem technologies. The innovative t echnology platform will include (i) a robot for the production of 2D protein crystals, (ii) the automated preparation of 2D crystals and native protein complexes for EM analysis, (iii) automated image acquisition and sample analysis by electron microscopy and (iv) management of all data produced in the pipeline. Each of these objectives represents a substantial advance of the state of the art. The innovations generated in HT-3DEM will greatly increase the throughput in 3D electron microscopy and thus enhan ce European competitiveness in the field of structural genomics. The reduction of processing times and cost for individual samples will open new fields to the application of this versatile technology platform. These are envisaged to include, among others, health care (screening of patient materials for infective agents) and pharma industry (acceleration of rational drug design). HT-3DEM thus responds to the needs of the European Community to improve human health and to increase the competitiveness of Europ ean pharma industry.
