Industrial Biotechnology is one of the Key Enabling Technologies identified by the Europe Union. Harnessing the power of microbes for biomanufacturing is one of the most promising approaches to providing a sustainable alternative to a fossil fuel- and animal farming-based economy. However, fermentation scale-up remains a massive challenge to the commercialization of biomanufacturing processes. At its core, fermentation scale-up involves bringing a biological process from an uncontrolled test-tube scale to a commercial scale of tens to hundreds of cubic meters. During scale-up, production output often fails to translate, undermining commercial potential and ultimately preventing the sector from reaching its potential.

Enduro has developed the game-changing synthetic-biology plugin Enduro Sense to bridge the scale-up gap and enable scalable and cheaper biomanufacturing. Our technology works to stabilize bioprocess strains to ensure that process output remains high throughout the scale-up process.

In the EU EIC Accelerator funded BioScale project, the aim was to showcase Enduro’s technology at industrial scale. During the BioScale project, Enduro successfully worked with industrial partners to implement the Enduro Sense technology in commercial bioprocess strains. The results confirmed the technology’s ability to significantly improve process stability and output in commercial operational environments. The results from the BioScale project cemented Enduro’s commercial potential and helped secure Series A investment.

During the BioScale project Enduro generated and screened hundreds of new strain candidates with the Enduro Sense technology. The technology implementation was carried across different organisms and product categories. By deploying iterative design-build-test-learn cycles, learnings were captured to improve the strain performance in subsequent implementation cycles. Many of the candidate strains showed an improved performance profile compared to the ancestral strain and were further validated in scale-down bioreactor operational environments. By the conclusion of the project, several industrially relevant Enduro Sense strains with significantly improved production stability and titer compared to the ancestral strains were identified and validated in bioreactors, highlighting the commercial potential of the Enduro Sense technology.
In the BioScale project Enduro’s Enduro Sense technology was demonstrated to stabilize bioproduction cells during bioprocess scale-up and scale-out, effectively bridging the scale-up gap. It was demonstrated that the technology can improve strain performance across organisms and products, highlighting to the universality of the technology and the broad marker relevance. With the successful demonstrations of Enduro’s technology, Enduro is poised to have a major impact on the biomanufacturing industry. The impact of the Enduro Sense technology on the biomanufacturing industry is centered around:
1) Increasing bioprocess titer and production rates with the same input for already established industrial strains.
2) Expediting commercial bioprocess development through predictable process scale-up.
The next phase, after the completion of the project, includes important communication of successes at industrial scale, which are set to be reached in the coming year.
