Directed Evolution of Enzymes that Modify Histone Tails

Objective

The modification of chromatin structure (e.g. by acetylation or methylation of the histone tails) plays a central role in transcriptional regulation, constituting an epigenetic code. Currently the dissection of the roles of individual modified histone residues is limited by low activity and/or specificities of isolated/recombinant histone modifying enzymes. We propose to combine directed evolution with biochemical and in vivo assays to understand the functional consequences of modifying specific histone residues. Natural histone-modifying enzymes will be used as starting points to evolve enzymes with have enhanced activities and altered specificities to probe biological function.
