Immunogenomics: Mouse to Human Translational Research

Final Report Summary - IMMUNO (Immunogenomics: Mouse to Human Translational Research)

IMMUNO funded the development of a high-throughput platform for detailed immune phenotyping of human samples, including the characterisation of 80+ immunological parameters from the peripheral blood. This information has now been compiled for more than 2500 samples, which includes healthy individuals, longitudinal samples, untreated autoimmune patients and autoimmune patients with a range of different treatments. The main dataset on healthy individuals (±1000 samples analysed) has unravelled key factors that influence our immune system, identifying the local environment as a key modifier of the healthy immune system. This dataset published as an open resource (Carr et al, Nature Immunology 2016), allowing the dataset to be assessed by the whole immunological community. In addition to studying the healthy context, IMMUNO assessed the immune system in a range of disease cases. We have assessed the impact of psychological factors on the immune system and the development of irritable bowel syndrome (IBS) (Wouters et al, Gut 2015). We have also
assessed the gene-immune relationship between changes that occur in Multiple Sclerosis (MS) and Autoimmune Thyroid Disease (AITD), and how these relationships are changed with treatment (Dooley et al, submitted). Finally, we have assessed several hundred pediatric cases of immune disorders, and have identified new gene-disease relationships (eg, Danso-Abeam et al, Orphanet Journal of Rare Diseases 2013; Van Eyck et al, JACI 2014; Van Eyck et al, Arthritis and Rheumatology 2015; Humblet-Baron et al, JACI 2016; Masters at al, Science Translational Medicine 2016; additional papers in preparation).
