Low-toxicity azine-based cationic lipids for gene transfection

Objectif

The project is aimed at the biochemical and biological investigation of a new class of molecules with azinic core (triazine, pyrimidine, etc.), featuring (1) detergent properties, for the transfer of oligonucleotidic genetic material (DNA, RNA, and mimics like PNA, etc.) into the cell nucleus (transfection), and (2) low cytotoxicity. The other principal structural features are: a thiol moiety for the formation of disulfide bridges in aqueous DNA and RNA, solutions; a protonable amine function not linked to the azinic core; a lipophilic chain to provide lipidic characteristics.These molecules could be used as transfection reagents for biological, biochemical and biomedicinal laboratories, and in perspective as drugs for gene therapy.The work will be carried o ut within the frame of a collaboration between the host Institution (CNR-ICRM) and Politecnico di Milano, which in turn hosts the CNR-ICRM section, in a new biology laboratory (Bio Cell) recently set up for the study of biological applications of engineeri ng and chemistry.
