Regulation of gene expression during liver development

Objectif

The main goal of this proposal is to understand the epigenetic mechanisms involved in the establishment and propagation of active chromatin states of genes during embryonic and postnatal liver development. The proposal is focused on a novel concept, put forward by the host group's previous studies, which suggest the generation of a poised, or committed state of active chromatin early during hepatocyte differentiation.

The research plan includes genome-wide gene expression analysis and biochemical approaches for chromatin structure and function studies in conjunction with DNA and RNA fluorescence in situ hybridization (FISH) assays. Besides wild type animals we also plan to study specific KO mice. While the host group has strong expertise in chromatin analysis using biochemical approaches it lacks the required experience in embryology, gene disruption and molecular imaging technologies. We seek to gain these latter expertise through the transfer of knowledge from recruited experienced researchers.

The host group will provide state of the art facilities and training in the field of gene expression and chromatin research. The project is expected to provide new insights into the poorly understood mechanisms involved in epigenetic regulation of active chromatin states. It will also have a great impact to the host in terms of enhanced competitiveness in the field of epigenetic research, technological competence and increased cohesion with European research.
