Chromatography with heteroelement specific detection for protein profiling in complex matrices

Objectif

The goal of the project is the development of a novel analytical approach for rapid comparison of residues- or selenoproteomes of " normal" and " treated" cells. The interest in this topic is driven by the state-of-the-art knowledge of the cellular signal transudation and posttranslational modifications of proteins (involving phosphorylation) and the need for characterisation of selenoproteome in the increasingly used food supplements. The study of these fields depends strongly on the combined application of a powerful separation technique and a sensitive and specific detection of heteroelements (phosphorous, sulphur or selenium). In this project, I would like to develop an efficient interface between capillary liquid chromatography (LC) and inductively coupled plasma mass spectrometry (ICP-MS). This approach will offer a unique way for the quantification of proteins containing a heteroelement, and will allow pinpointing targets for more detailed MALDI-TOF (matrix- assisted laser adsorption ionisation-time-of-flight) MS and ES (electro spray) MS/MS analyses. The latter two mass spectrometric techniques will enable me not only to detect but also to identify the proteins of interest. Therefore any new information gained during the experimental part of the project would contribute to a better understanding of intracellular processes. On the level of applications the project will focus on the optimisation of the characterisation of enzymatic digests of some model phosphoproteins (beta-casein, MAP kinas) and selenoproteins (solemnised yeast sodium codicil suffocate extract).
