Practice abstracts for BCAs, plant resistance, improved BCA strains and Integrated Pest Management

"The resulting innovative knowledge and easy accessible end-user material from this project will feed into the EIP-AGRI (The agricultural European Innovation Partnership) website for broad dissemination. The end-user material to be produced contains a substantial number of summaries for practitioners in the EIP common format (""practice abstracts""), including the characteristics of the project (e.g. contact details of partners, etc). A full package of practice abstracts will be produced by the project, containing all the outcomes/recommendations which are ready for practice.A ""practice abstract"" is a short summary of around 1000-1500 characters (word count – no spaces; see below) which describes the main information/recommendation/practice that can serve the end-users in their daily practice. Guidance and templates for these practice abstracts are available on the EIP-AGRI web site: http://ec.europa.eu/eip/agriculture/en/content/eip-agri-common-format. A total target number of 4 practice abstracts is foreseen for the project. Practice abstracts (PA):1. BCAs kai plant resistance kai improved BCA strains2. Integrated Pest Management (one system)"

Manuscript on tomato interactor proteins and their binding domains including the effect of S-gene knock-down in tomatoes

To identify targets of P1 and D1, we will perform pull down on tomato protein extracts in collaboration with Geert de Jaeger (VIB, Belgium). Additionally, recombinant protein will be generated of P1 and D1 fused to a STREP-tag. After isolation, the proteins will be bound to anti-STREP Sepharose beads and incubated with tomato phloem exudates from control and whitefly-infested plants. The bound proteins will be subjected to mass-spectrometry for the identification of interactors. Interactors will be confirmed using yeast-2-hybrid (Y2H), transient bimolecular fluorescence (BiFC) assays and co-immunoprecipitation (co-IP). We will investigate whether these confirmed tomato targets are indeed used by whiteflies to improve the hosts’ suitability. The following criteria will be considered to decide which tomato targets to study in detail

Protocol for predicting the effect size of a new resistance on prey equilibrium densities using predator-prey models
Report on the variability and adaptation after selection to tomato among wild populations of the biological control agent O. laevigatus and A. swirskii
Report for available screening platforms and pipelines

The integrated and fully automated diagnostic platform will be validated against a number of different samples from several geographical regions in comparison to existing gold standard assays This concept has a high probability of success because the disc cartridge has been tested in other systems eg human diagnostics It is robust highly sensitive and specific and can be very adaptable to diagnostic needs in terms of the nature and number of detectable species including applications related to vector control ie wwwdmcmalveceu The LabDisk will offer higher added value compared to performing individual assays as it will also include markers for metabolic resistance and provide sufficient information for operational level decision making at an affordable cost

Manuscripts on functional characterisation of resistance (target: >2)

The results will be published in scientific journals and conferences

Manuscript on the salivary effector proteins of whiteflies.

Collect and sequence salivary proteome from B tabaci on artificial diet mapping references are available select putative effector candidates transiently overexpress top 10 candidates in tobacco and assess the impact on whitefly performance Select the best effector candidate

Manuscript on biochemistry of plant breeding based on elicitors / R-genes and effectors / S-genes of whiteflies and other Superpests on crops

Knock out and knock down of maximally three S-gene targets, determine the site where the S-protein interacts with the effector in the S-protein, generate MicroTom with mutated S-gene, assess the life tables of MicroTom with mutated S-genes.

The novel molecular markers that will be included to the LabDisk in order to have fast and reliable results with low cost

Novel molecular markers for emerging resistance cases (target: >5)

We will investigate field populations sampled from various economically important crops that are reported by farmers and extension services to be particularly difficult to control with current chemical strategies, implying the evolution of resistance. We will prioritise cases of potent and novel resistance (i.e. chemistry that has not been previously compromised by resistance). As a consortium we are part of, or have access to, extensive monitoring networks, which will provide access to these resistant populations. The following compounds/pests will be included in the analysis: tetronic acids (keto-enols) – spirodiclofen, spiromesifen and spirotetramat / T. urticae, B. tabaci and M. persicae; pyflubumide, cyenopyrafen and cyflumetofen (most recently developed acaricides acting on Complex II of the mitochondrial electron transport chain, recently introduced to the market) / T. urticae; abamectin (a neurotoxic insecticide/acaricide of very low toxicity to mammals) / F. occidentalis, T. urticae; spinosad (a low toxicity insecticide of natural origin) / F. occidentalis; pymetrozine/flonicamid / B. tabaci and M. persicae.
