Objectif
The use of hemopoietic growth factors for irradiation therapy have been investigated. Although a relationship between the early GM-CSF response and the dose of irradiation could be established, the variability of response and slow response time made use of GM-CSF impractical. However, GM-CSF was totally effective against leukopenia after a radiation dose of 5 Gy total body irradiation (about 2-log stem cell depletion), but it became increasingly less effective at higher doses of irradiation. Studies with the much more broadly acting growth factor IL-3, were also carried out. Molecular cloning and production of rhesus monkey IL-3 was successfully achieved and preclinical pilot studies, using unirradiated monkeys, have been completed. The positive selection of hemopoietic stem cells was also achieved, by developing a large-scale method based on selection of CD34+ cells. These preparations have been successfully tested in autologous transplantation experiments.
