Exploitable results

The project has provided - basic knowledge about the production of polymer based particles useful for enzyme immobilisation and for the purification of proteins, - prototypes of matrixes with direct potential as enzyme carriers, - identification of novel immobilization methods and techniques, - basic knowledge about the possibilities and limitations of using immobilized enzymes in food technology and other areas where commercially available enzymes today are used on an industrial scale. In one task of the project antibody fragments with different specificities and affinities for a drug racemate were obtained by cloning and directed site-specific mutagenesis. Different immobilization tags were introduced in order to immobilize the antibody fragments. The antibody fragments were produced in E. coli and purified. Antibody fragments with excellent specificity in separating enantiomers could be produced. Site-specific mutagenesis was used to lower the affinity if needed and to improve the reusability of antibody containing affinity columns. Thus, the concept of using antibody fragments for enantiospecific chromatographic separation of organic molecules was proven to be successful. These types of immunoaffinity columns have a high potential especially as an analytical tool for separating closely related small molecules of drug, environmental or food origin.identi
