Objectif DEVELOPMENT AND VALIDATION OF ASSAYS TO EVALUATE CHEMICALS CAPABLE OF INDUCING GENOMIC MUTATIONS. EMPHASIS IS GIVEN TO THE FUNGUS ASPERGILLUS NIDULANS, WHICH OFFERS CONSIDERABLE PROMISE AS A SYSTEM FOR DETECTING GENOMIC MUTATIONS AND FOR UNDERSTANDING THE MECHANISMS OF SUCH MUTATION. THE WORK INCLUDES : I) THE USE OF THE DIPLOID STRAIN OF A NIDULANS TO DETECT CHEMICALLY INDUCED EUPLOID MITOTIC SEGREGANTS AS PRODUCTS OF NON-DISJUNCTION OR MITOTIC CROSSING-OVER. II) THE SYNTHESIS OF MARKED DIPLOID STRAINS TO DISTINGUISH INDUCED ANEUPLOIDY FROM CHROMOSOME BREAKS. III) INVESTIGATING THE POSSIBILITY OF USING METABOLIC ACTIVATION IN THE SYSTEM. IV) ESTABLISHING THE EFFECTS OF ANEUGENIC CHEMICALS ON TUBULIN ASSEMBLY.
