PROCESSING TECHNOLOGY FOR RECOVERY OF RECOMBINANT ANTIBODY PRODUCED IN CROP PLANTS

Objectif

The project is concerned with the discovery and design of stable peptides as biding ligands for antibodies and to produce stable affinity media for the efficient purification
of recombinant antibody produced in plants.

The intention is not to use whole or even fragmented antigen on the affinity surface but rather to use small stable peptides which mimic the natural epitope recognised by the antibody. The ideal
ligand will allow controlled capture and release during processing thereby yielding purified active antibody.

The project focuses on this extraction process because the effectiveness of downstream activities are crucial to the vision of low cost production of antibody fragments as fine chemicals produced by agricultural technology.

The objectives of the project are :

- to develop and scale-up antibody purification processes that exploit the
antibody's inherent ability to bind its epitope, for maximum recovery of active
material from plant tissues;
- to devise specific affinity purification media, based on peptide antigen analogues
designed to capture and release antibodies, in a stable and multiple re-useable
system;
- to transfect genes of appropriately designed antibodies into tobacco and potato
(production organisms), to provide material on a practical scale for process
development;
- to develop process control and product QC techniques to quantify, monitor and
validate the production and purification processes;
- to integrate plant culture and harvesting with tissue extraction and affinity
processing to develop and define a scale-up pilot process;
- to evaluate the performance characteristics of the working pilot system in terms
of active product yield, reusability/stability of immobilised peptide, cost efficiency
and ease of further scale-up.
