Distribution of allergens and allergen-coding mrnas in olea europaea tissues

Objectif



Olive pollen is considered to be one of the major causes of respiratory allergy in Mediterranean areas. Recent research has identified and purified major olive allergens at the biochemical level and several programmes have already been initiated in order to determine the ultrastructural localization as well as to clone olive allergens.
During the tenure of the grant it is intended to determine the distribution of allergens and allergen-coding mRNAs in different olive tissues and at different stages of development. The method will be based on in vitro translation of mRNAs extracted from the tissues to be tested. The translated proteins will be analysed by SDS-PAGE, transferred to membranes and probed using a variety of sera and MAb to allergens.
The goal of the project is to analyse the spatial and temporal replatio of the expression of allergen genes, a crucial step towards understanding their function and their eventual manipulation. New data about cross-reaction between allergenic proteins in different species are also expected.
