Structural and functional analysis of nup84, a novel nuclear pore complex protein associated with can/nup214

Objective



The interphase nuclear envelope is a selective barrier which defines the boundary between the nucleus and cytoplasm and as such governs the transport of macromolecules between these two compartments. In this regard the nuclear envelope plays a central role in determining nuclear activity during cell growth and development. The channels which mediate this regulated traffic of molecules across the nuclear envelope are the nuclear pore complexes (NPCs). These are not simple pores embedded in the nuclear envelope but large and elaborated supra molecular assemblies. At present the nature of the molecular machinery within the pore complex and interactions with other nuclear and cytoplasmic components remain largely obscure.
We have characterized a group of nuclear complex proteins and begin to map their functional and physical interactions. It is the goal of this proposal to study in greater detail a novel component of the NPC that we have identified and cloned. This will provide new information on the organization and function of the NPC.
