Objectif Proliferation is one of the physiological processes regulated by cAMP. The cAMP action mechanism involved variations in gene expression. CREM gene is one of cAMP effectors at nuclear level that has been discovered in the last years. Evidence have been accumulated that indicate a possible role of CREM in regulation of cell proliferation. In particular it is significant that CREM proteins regulate protooncogene c-fos and cyclin A gene expression, and their function is controlled by important cell cycle regulators such as p70S6 kinase and the antioncogene p53. The research project proposed aims at improving the actual knowledge on transcription factor CREM and specifically on his role in the regulation of cellular proliferation and transformation. The approach consists in a series of different experiments to test directly CREM transforming properties and its antioncogene ability, through increased expression or specific elimination of different CREM proteins both in vitro and in vivo.
