Chemo-enzymatic synthesis of complex carbohydrates - discovery of new enzymes using fluorescence-tagged substrates and fourier transform ion cyclotron resonance mass spectrometry

Research objectives and content
The objectives of the research new glycosidases with specificity for the creation of B1->3, 1->4, or or 1->6 linkages to lactosamine derivatives from N-acetylglucosamine to create linkage types of importance in glycobiology. An approach combining detection of specific hydrolytic activity towards the target Resonance Mass Spectrometry (FTICRMS) will be developed. Enzymes detected by the hydrolytic mode of screening will then be tested for their synthetic capabilities, again using
fluorescence-tagged substrates with FTICR-MS confirmation of product structures.
Training content (objective, benefit and expected impact)
I will receive training in novel assay methods for the discovery of glycosidases useful for the synthesis of complex carbohydrates. The project will also give me experience in conventional chemical methods for oligosaccharide synthesis and in many advanced analytical techniques, particularly FTICR-MS.
Links with industry / industrial relevance (22)
The project will be of great interest to workers in glycobiology interested in the role of the glycosyl components of glycoproteins, many of which are in industrial laboratories. The host group at Warwick has extensive links with industry.
