Objectif
The proposal is aimed to contribute to understanding of pathophysiological and seroepidemiological aspects of water buffalo fasciolosis, to analyse the usefulness of classical and new diagnostic methods and to study the efficacy of new therapeutic approaches.
The main objectives are:
* To study biochemical markers of the disease in acute, subacute and chronically infected animals.
* To evaluate modifications in the synthesis of inflammatory mediators of eicosanoid type in infected animals and to correlate these changes with clinical manifestations of the disease.
* To analyse changes induced by Fasciola spp. in liver metabolism by means of antipyrine saliva clearance and study of its urinary metabolites.
* To standardise ELISA techniques using ES products from Fasciola spp. for the serodiagnosis of water buffalo fasciolosis.
* To examine the prevalence and distribution of water buffalo fasciolosis in Jiangsu and Anhui Provinces (China) using both the standard coprological examination and the ELISA techniques.
* To standardise a PCR technique for the detection of Fasciola spp. stages in the intermediate hosts.
* To study patterns of food and water intake in water buffaloes and the changes induced by Fasciola spp. infection.
* To analyse the mechanisms of food intake depression in fasciolosis by selective denervation of infected animals.
* To develop slow-release boluses of drugs already used against Fasciola spp and to analyse the efficacy of these new formulations.
Expected Outcome
The work carried out should provide knowledge concerning the pathophysiological aspects of water buffalo fasciolosis and the identification of biochemical and serological markers that could be correlated to the different stages of the parasitosis and used to examine its prevalence, distribution and relation to the use of anthelmintic programs. The development of ELISA and PCR techniques for detection of parasite stages in the final or intermediate host would offer completely new tools for routine diagnosis and epidemiological surveys. The development of slow-release boluses of drugs would suppose novel therapeutic approaches. The scientific qualification of the European partners guarantees the training value of the proposal. Chinese researchers that keep a permanent link with their institutions will benefit from research experience in EU laboratories and from the help of European scientists visiting China. The transmission of this experience and knowledge to graduate and undergraduate students will extend the research experience and the efficiency of RTD utilisation in developing country situations.

* Study of biochemical markers of the disease in acute, subacute and chronically infected animals.
* Determination of the eicosanoid profile in blood during the development of the parasite.
* Analysis of changes induced by Fasciola spp. in liver metabolism by means of antipyrine kinetics.
* Standardisation of an ELISA technique for the serodiagnosis of water buffalo fasciolosis.
* Detection of Fasciola spp. stages in the intermediate hosts by a PCR technique.
* Study of the patterns of food and water intake in water buffaloes and analysis of the mechanisms of food intake depression induced by Fasciola spp.
* Development of slow-release boluses of drugs used against Fasciola spp.
