Production of valuable breeding material of eggplant (Solanum Melongena L.) resistant to fungal and bacterial wilts, and root knot nematodes by using protoplast fusion

Objectif

The main objectives are defined as follows :
* To overcome Fusarium and Verticillium wilts, Pseudomonas wilt and nematode attack by the production of healthy and resistant breeding material of eggplant (Solanum melongena) with reduced use of pesticide and fertilizer for a better preservation of environment.
* To use biotechnological methods, particularly including protoplast fusion, to supplement and complement conventional breeding methods of eggplant with valuable breeding lines carrying traits of resistance.
* To exploit fusion products and to promote the selected hybrids for their potential use as valuable breeding material.
Expected Outcome

In this project, new techniques, particularly protoplast fusion, anther culture, flow cytometry for DNA analysis, and molecular techniques, will be adapted to the production and the exploitation of highly valuable breeding lines of eggplant with resistance to bacterial and fungal wilts and nematodes. This will supplement and complement the conventional breeding methods of this popular vegetable crop. Fundamental knowledge about the potential transmission of disease resistance characters, the cellular and organism response to pathogens and inter-genomic recombination, as well as the agronomic value of breeding lines obtained is expected as direct outcome of the project. In addition, breeding materials with high agronomic value and pathogen resistance will be produced and utilised in eggplant breeding programmes.
The key activities involve :
* Assessment of traits of resistance to Fusarium and Verticillium wilts, Pseudomonas wilt and root knot nematodes in different accessions of Solanum aethiopicum, S. integrifolium, S. sisymbriifolium and S. torvum in view of their use as fusion partners. Accessions of those wild relatives from China, India and Indonesia, as well as those from the collection of the Istituto Sperimentale per l'Orticoltura (Italy) and the Institut National de la Recherche Agronomique (France) will be evaluated for resistance. The genotypes with high level of resistance will be characterised and selected as fusion partners.
* Production and characterisation of interspecific somatic hybrids of eggplant by using protoplast fusion. Both the techniques of electrofusion and PEG-mediated fusion will be optimized and adapted to the production of somatic hybrids and asymmetric hybrids of eggplant. Somatic hybrids and asymmetric hybrids will be characterized morphologically, cytologically (classical cytology and flow cytometry) and biochemically (isozymes, RAPD analysis and RFLPs).
* Exploitation of fusion products and promotion of the selected hybrids for their potential use as valuable breeding material. The allotetraploid somatic hybrids and asymmetric hybrids will be evaluated for resistance and hybrid fertility in particular. Fertile 4x hybrids will be brought back to the diploid status by using the technique of anther culture. Both 4x somatic hybrids and 2x-anther-derived plants, as well as asymmetric hybrids will be backcrossed to the cultivated eggplant. The resulting progenies will be evaluated for hybrid fertility, resistance, agronomic value and inter-genomic recombination. The breeding lines with high level of resistance and agronomic value will be selected, and released to farmers and breeders for practical evaluation and the promotion of their potential usefulness in eggplant breeding programmes.
