From conopeptide scaffolds to analgesic leads

Objective

This proposal is focused on the use of conopeptide scaffolds for discovery and engineering of novel analgesics. A combination of polemic and selection methodologies will be used to identify conopeptide legends for receptors of clinical importance in chronic and naturopathic pain syndromes, namely the SNS tetrodotoxin-resistant sodium channel, and the p75 neurotrophin receptor. The multidisciplinary research program combines efforts of seven academic and two industrial groups, working in fields that include protein chemistry, protein folding and engineering, functional genomic, interaction selection systems, molecular physiology of pain pathways, and CNS delivery mechanisms.
