Risk assessment of wood dust: assessment of exposure, health effects and biological mechanisms

Objective

The goal of this proposal is to provide accurate, up-to-date data on occupational exposure to wood dust, and to assess associations between such exposure and molecular alterations of Sino-nasal cancers, and biological mechanisms of pulmonary inflammation. Data on overall levels of European exposure will be obtained from European exposure databases and exposure registers as well as by monitoring wood dust levels in workplaces. Sino-nasal cancer cases and their exposure histories will be collected and paraffin-embedded tissue samples examined for p53 and K-transmutations. DNA damage induced by wood dust will be studied in exposed human cells. Molecular mechanisms of wood dust-induced pulmonary inflammation will be studied with experimental animals and cell cultures, and with nasal epithelial cells obtained from wood dust exposed subjects. Data from exposure assessment and the toxicological endpoints will be compiled for risk assessment. The study ultimately aims to provide exposure and mechanism-bases data for setting occupational limit values to improve safety at workplaces.
