Production and field evaluation of healthy micropropagated sweet potato. Development of resistant cultivars to virs complex by plant tissue culture (somaclonal variation and protoplast fusion)

Objective



THE OBJECTIVES OF THIS PROJECT ARE TO EVALUATE DIFFERENT STRATEGIES 0F CONTROL OF
BACTERIAL WILT AND VIRUS DISEASES OF SWEET POTATO IN THE PROVINCE 0F GUANGDONG (CHINA) AND THE STATE OF RIO DE JANEIRO (BRAZIL).
IN THE SHORT RANGE, OUR PROJECT AIMS AT :
-PROVIDING A STEADY SUPPLY OF INDEXED MATERIAL THROUGH IN VITRO CULTURE FIED PROPAGATION (BRAZIL AND CHINA) AND TO ANALYSE THE RATE OF REINFECTION -IMPROVING DIAGNOSIS 0F THE DIFFERENT COMPONENTS OF THE VIRUS DISEASE COMPLEXES.
PURIFIED VIRUS WILL BE USED TO PREPARE POLYCLONAL ANTISERA WHICH WILL BE EVALUATED BY ELISA TESTS.
IN A LONGER RANGE STRATEGY, OUR PROJECT AIMS :
- THE BEST CULTIVARS OF SWEET POTATO FROM BRAZIL, CHINA,INDONESIA WILL BE INTRODUCED
INTO CELL AND PROTOPLAST CULTURE, IN ORDER TO SELECT AND IMPROVE EXISTING VARIETIES BY SOMACLONAL VARIATION. (FRRENCH PARTNER, ORSAY). REGENERANTS WILL BE TESTED UNDER THE FIELD CONDITIONS IN BRAZIL AND CHINA IN ORDER TO IDENTIFY POTENTIALLY USEFUL SOMACLONAL VARIANTS FOR THEIR RESISTANCE TO VIRUSES, AND TO BACTERIAL WILT IN CHINA.

- THE CHARACTERIZATION oF GENOTYPES 0F SWEET POTATO CLONES (FRENCH AND INDONESIAN PARTNERS) BY THE TECHNIQUES OF ELECTROPHORESIS OF ISOENZYMES. - THE CHINESE PARTNER WILL CONTINUE TO ADJUST THE PARAMETERS 0F IN VITRO STORAGE UNDER GROWTH LIMITING CONDITIONS.

MEANWHILE, THE ENGLISH PARTNER WILL BE IN CHARGE OF THE STUDY 0F CRYOCONSERVATION OF SWEET POTATO GERMPLASM, USING THE BEST CURRENTLY AVAIBLE CHINESE AND BRAZILIAN CLONES AS MODEL.
