STUDIES OF PROLIFERATIVE KIDNEY DISEASE WITH PARTICULAR REFERENCE TO PROPHYLAXIS AND GEOGRAPHICAL DISTRIBUTION IN EUROPE

The object of the study is to use specific probes to identify the surface antigens and life cycle stages of PK"X", the causative agent of PKD, so that an effective vaccine against the disease can be developed. In addition, the distribution and level of the disease in Europe will be established. Monoclonal antibody and lectin probes will be used to identify and quantify the parasite in fish tissues and water samples collected during the PKD season throughout the course of the project. Monoclonal antibody probes, developed during the "FAR" project, will be used to purify PK"X" cell antigen from host cells using affinity chromatography. These will be used to inject mice and produce a further battery of monoclonal antibodies to different surface antigens. Such probes will be used in life-cycle studies and enable identification of life-cycle stages prior to those currently identified using histological methods. The detailed immune response of fish following natural PKD infection will be investigated. ELISA and dot-filter tests will also be used to detect specific PK"X" cells from water samples by linking monoclonal antibodies to filters. Such material will then be used in attempts at culture of the PK"X" cells in vitro. A series of passive immunisation experiments will be carried out to establish the nature of the protective antigens. This information will then be used to formulate and field test potential vaccines.
