During the project, a comprehensive set of technical and scientific activities was successfully completed. The therapeutic potential of MDC-735 was first validated in vitro using cell killing assays, which demonstrated high sensitivity of both platinum-sensitive and platinum-resistant ovarian cancer cells to MDC-735 treatment. Moreover, the potential of MDC-735 to infilitrated ovarian cancer tumoroids in vitro has been confirmed as well as killimg cancer cells. Additionally, the induction of phagocytosis and immune activation has been confirmed after treatemtn with MDC-735.
Subsequently, the efficacy of MDC-735 was evaluated in multiple in vivo ovarian cancer models. In mouse models implanted with platinum-sensitive as well as platinum-resistant ovarian cancer cells, treatment with MDC-735 resulted in a significant extension of survival compared to control groups. Most importantly, in models of platinum-resistant ovarian cancer, MDC-735 treatment led to complete tumor eradication in approximately half of the treated animals, representing a highly exceptional outcome in this disease setting. MDC-735 confirmed its efficacy in PDX platinum-resistant ovarian cancer models.
In parallel, supporting preclinical studies were conducted to characterise the biodistribution and biological behaviour of the drug delivered via macrophages, providing essential data for defining dosing strategies and therapeutic windows. The dose finding and dose-dependent efficacy staudy were also preformed. Toxicology study confirmed safety of MDC-735 drug product. Together, these activities delivered a robust and coherent preclinical efficacy package demonstrating the feasibility and therapeutic promise of this novel cell-based approach.
In parallel with the preclinical development activities, the project also addressed key translational aspects required for further development and exploitation. A preliminary business case and development scenarios were prepared, taking into account different pathways towards clinical development, partnering and future commercialisation of the technology. These activities contributed to assessing the long-term viability and strategic positioning of the MDC-735 product.
Furthermore, intellectual property arising from the project results was consolidated. A patent application covering key aspects of the MDC-735 technology was prepared and filed, ensuring protection of the core innovation and strengthening the foundation for future exploitation and investment.
Together, these activities resulted in a coherent and mature preclinical and translational package, positioning MDC-735 for the next stages of development towards clinical application.
