Single-molecule Nanopore Analysis Platform for RNA Readout

Objective

RNA viruses, including Zika, SARS-CoV-2, and Hepatitis C, impose a profound global health burden, causing millions of deaths every year. The extraordinary rates of mutations and chemical modifications in these RNA-based pathogens enable rapid adaptation and, ultimately, immune evasion. Current detection technologies fail to capture the molecular heterogeneity that drives viral evolution, relying on ensemble averaging, biased enzymatic amplification, or computational inference to compensate for low signal-to-noise. SNAPRR aims to overcome these limitations by developing a unified single-molecule platform that identifies viral RNAs and their underlying mutations, and chemical modifications, revealing the hidden chemical diversity that shapes infectivity and pathogenesis.

SNAPRR harnesses three emerging technologies to sensitively and specifically probe viral RNA chemistry. 1) Solid-state nanopores translate individual RNA molecules into recognisable ionic current signatures, enabling rapid, quantitative readout. 2) Programmable nucleic acid nanotechnology uniquely barcodes viral RNAs for multiplexed detection, and 3) Chemoselective labelling amplifies signals from single-nucleotide mutations and chemical modifications for direct nanopore localisation.

By revealing strand-to-strand heterogeneity, SNAPRR will redefine our understanding of RNA virus biology, inform next-generation diagnostics, and provide a versatile framework for antiviral development and RNA vaccine quality control. Bridging chemistry, nanotechnology, and biophysics, this project combines fundamental insight with practical impact, creating a portable, amplification-free approach to tackle the global challenge of rapidly evolving RNA viruses.

Fields of science (EuroSciVoc)

CORDIS classifies projects with EuroSciVoc, a multilingual taxonomy of fields of science, through a semi-automatic process based on NLP techniques. See: The European Science Vocabulary.

Keywords

Project’s keywords as indicated by the project coordinator. Not to be confused with the EuroSciVoc taxonomy (Fields of science)

Programme(s)

Multi-annual funding programmes that define the EU’s priorities for research and innovation.

Topic(s)

Calls for proposals are divided into topics. A topic defines a specific subject or area for which applicants can submit proposals. The description of a topic comprises its specific scope and the expected impact of the funded project.

Funding Scheme

Funding scheme (or “Type of Action”) inside a programme with common features. It specifies: the scope of what is funded; the reimbursement rate; specific evaluation criteria to qualify for funding; and the use of simplified forms of costs like lump sums.

HORIZON-ERC - HORIZON ERC Grants

Call for proposal

Procedure for inviting applicants to submit project proposals, with the aim of receiving EU funding.

Net EU contribution

Net EU financial contribution. The sum of money that the participant receives, deducted by the EU contribution to its linked third party. It considers the distribution of the EU financial contribution between direct beneficiaries of the project and other types of participants, like third-party participants.
