Final Report Summary - PARIS (PHYSICIAN AIDED RECONSTITUTION OF THE IMMUNE SYSTEM)

Our goal has been to manipulate the lymphocytes of the immune system following their therapeutic depletion with antibodies so as to enhance their ability to regulate or dampen immunity, rather thanto evoke it. If this could be done with short-term therapy it would provide a valuable alternative to
long-term immunosuppression. In the PARIS programme we have sought ways to enable FoxP3+ regulatory T-cells (Treg) to become functionally dominant in the reconstitution period. This far we have defined the extent to which different lymphocyte growth cytokines can contribute
to that goal, and using novel state of the art flow imaging, established which conditions allow regulatory T-cells to proliferate, and have provided clues as to how to selectively promote Treg reconstitution during homeostatic expansion by PARIS after therapeutic lymphocyte ablation.
