Objetivo The aims of this proposal are to study how lymphatic vessels grow in health and disease, and to identify and characterize novel candidate genes of lymphangiogenesis important for human disease and possible targets for drug development of lymphatic disorder s. This will be achieved by performing functional lymphangiogenomics. First by using Xenopus laevis tadpoles, a novel model organism to on a large scale and rapid pace study lymphangiogenesis, the function of genes of interest (selected by micro-array profiling) will be studied via genetic manipulation (RNA/DNA over expression and morpholino knockdown).The most promising lymphangiogenic candidates will then be knocked down in the mouse and their role in health and disease will be studied as well as their molecular mechanisms in vitro and the therapeutic potential for treating lymphatic disorders in vivo.
