RESPONSE TO RADIATIONS OF HUMAN CELLS MODIFIED BY pR PLASMID THAT CONFERS RADIORESISTANCE IN BACTERIA

Obiettivo

EVALUATION OF GENETIC RISKS FROM IONIZING RADIATION. DETERMINATION OF CELLULAR RADIOSENSITIVITY AND RELATION TO REPAIR. THE AIM OF THE PROPOSED RESEARCH PROJECT IS TO STUDY THE RESPONSES IN TERMS OF SURVIVAL, REPAIR OF DNA AND ANALYSIS OF INDUCED DAMAGE IN HUMAN CELLS MODIFIED BY PR PLASMID AFTER TREATMENT WITH UV RADIATION AND UV MIMETIC AGENTS.
The purpose of this research program is:
to clarify the function of the uvp1 and uvp2 (mucAB genes) regions of pR plasmid;
to investigate the interaction between them;
to study the possible correlation between their expression in normal and defective repair pathways in mammalian cells.
The main objectives are:
the molecular characterisation of the pR uvp1 region in terms of nucleotide sequences and codified protein in bacterial cells;
the analysis of pR product interactions with mutagenesis and with the different repair pathways in bacterial and mammalian cells;
the characterisation of the pR effects in radiation sensitive mutant cells, such as ataxia telangiectasia cells, through survival analysis and chromosome damage evaluation.

The radiation response of human cells modified by pR plasmid, which confers radiation resistance to bacteria, has been studied.
The main objectives were:

THE STUDY OF THE UV RADIATION RESISTANCE CONFERRED ON MOUSE AND HUMAN CELLS TRANSFECTED WITH THE PR PLASMID WHICH CARRIES UV+ GENES. CELL SURVIVAL WILL BE MEASURED TOGETHER WITH THE DETERMINATION OF DNA REPAIR AND THE EVALUATION OF THE INDUCTION OF RECOMBINOGENIC EVENTS GIVING CHROMOSOMAL ABERRATIONS.
