Development, optimization and validation of molecular tools for assessment of biodiversity in forest trees

Management of biodiversity in forest trees is focused on stabilization of highly diverse, utilised ecosystems with species of extraordinarily high genetic diversity.
Molecular methods are needed as prerequisite for monitoring genetic diversity. As a joint effort of both producers of molecular technologies and representatives of technology users, the project will contribute to the further development of rapid and effective molecular methodologies which are valid for a broad range of end users. The methodologies will help to implement the Rio / Strasbourg / Helsinki process in conservation of biodiversity of European forests.
The proposal is based on the achievements of a biodiversity project in Framework III and aims at exploiting this knowledge by the optimization and validation of tools and making them available for the end users' objectives. As a result of FRW III, PCR-based technics were evaluated as most promising key technology. Thus the main efforts are focused on the characterization of sequences and primer identification. Oak and Norway spruce are chosen as model species.
In line with the research tasks needed to reach the project goals two working packages (WPs) are proposed. WP `Development of methodologies' consists of the activities `Optimization of technologies', `Development of universal tools, and `Sampling strategies and data handling'. WP `Validation of methodologies' consists of the activities `Tests on reproducibility, universality and efficiency', `classification of markers (which marker for which purpose), and `transferability of technologies to end users'.
Main project milestones and deliverables are DNA banks, technical manual for automation, (primer) sequences and cDNA libraries, genetic maps, manual of data handling and software for inheritance analysis, outline of applicability of which marker to which taxonomic range, compendium `which marker for which purpose', tradeoffs on markers, and report on transferability. The partnership consists of 14 participants, representing different complementary kinds of technology producers and technology users of various categories. For implementing the WPs, the tasks and milestones as mentioned above and certain service functions are allocated to specific responsible participants.
Main address of the project exploitation are the end users in the field of biodiversity in forestry. The International Union of Forest Research Organizations (IUFRO) and a platform on `Biotechnology for Biodiversity' function as an extended audience for dissemination of the results.
