Induced Resistance of Plants to Pathogens: Triggering and Expression

Plants can be induced to develop broad spectrum resistance which is effective against a wide range of pathogenic microbes, however, the exploitation of this phenomenon in crop protection is currently hampered because of our ignorance of the mechanisms involved. A number of distinct induced resistance mechanisms exist in plants which can be activated, locally and/or systemically, in response to pathogenic and non-pathogenic microorganisms.
The objective of this proposal is to understand the molecular and physiological basis of a set of induced resistance phenomena termed systemic acquired resistance (SAR), induced systemic resistance (ISR) and localised induced resistance (LIR). Although SAR, ISR and LIR have distinctive features there are also similarities, suggesting that at least some of the pathways governing these responses share common components.
The participants will carry out a concerted programme of complementary genetic, molecular biological, biochemical, cytological and pathological experiments, using both the model plant Arabidopsis and a selection of crop plants. Resistance will be induced with various microbes and fractions therefrom and induced plants will be challenged with a range of fungal and bacterial pathogens. In addition to laboratory experiments, realistic greenhouse tests will evaluate generality and practical utility.
The research will be divided into three Work Packages. Work Package 1 will be concerned with the nature of the microbial components inducing resistance and their perception by the plant. The major strategies to be used will be biochemical fractionation and characterization of inducer-active material, together with microbial genetic approaches (mutants altered in inducing ability, cloning of cognate genes). Work Package 2 will be concerned with the mechanisms by which plants transduce the perceived microbial signals. The major strategy will be genetic (isolation and characterization of Arabidopsis mutants), supplemented with appropriated biochemical and cytological studies. Work Package 3 will be concerned with the expression and mechanisms of induced resistance, including the nature of the antimicrobial state or substances. Strategies will include both microbial and plant genetic approaches (mutants and study of gene expression) and biochemical analysis.
Exchanges of information and biomaterials among participants will allow unprecedented comparison of the diverse features of induced resistance. The programme will permit evaluation of the generality of the phenomena among plants and microbes and assessment of the feasibility of large scale exploitation as a component of environmentally-friendly crop protection strategies.
